The Eastern States 100 is a single-loop ultramarathon located in the Pennsylvania Wilds. The course covers a distance of 103 miles and circumnavigates the Pine Creek watershed. Participants navigate a route that includes over 20,000 feet of total vertical climbing on technical trails.
The event is held in Waterville, Pennsylvania, and utilizes the various trail systems surrounding the Pine Creek Valley. The race route is designed as a full circumnavigation of the Lost and Bewildered River area.

The event is managed by the Eastern States Trail-Endurance Alliance, an all-volunteer grassroots organization. The course features multiple aid stations for participant support. Recent updates to the course include the relocation of Aid Station 2 to Bonnell Flats, situated at mile 11.6 along the Pine Creek Rail Trail. Due to the remote nature of the course, certain locations such as the Barrens aid station (AS14) have restricted access where crews are no longer permitted.
