The Conestoga Trail Run is an established trail race in Lancaster County, Pennsylvania, that has been held for over 30 years. The course covers 10 miles of technical terrain characterized by steep hills, various rock formations, and multiple small stream crossings. Runners navigate through scenic landscapes featuring views of the Susquehanna River, clear feeder streams, and native flora such as Mountain Laurel and Rhododendrons.

The race follows a point-to-point format. Participant check-in and bib pickup take place at Kellys Run Nature Preserve in Holtwood. From there, the event provides one-way bus transportation to the starting line at Pequea Creek Campground. The finish line is located back at Kellys Run Nature Preserve.

The course is marked with orange ribbons and includes sections on both public trails and private property. There is a strict time limit for this event; participants must reach the Pinnacle Aid Station at mile 7 within 2 hours and 30 minutes to continue. Due to the technical nature of the trails and potential environmental factors like bees, the event is intended for experienced trail runners.
