The Perry Lake trail for the 50K loop is divided into five sections and includes about 3,300 feet of elevation gain. The route is marked with blue paint on trees and posts; reflective blue arrows and double blue marks indicate turns. Markings are expected to be visible in December.

The first section leaves Apple Valley, climbs through Longview Park, skirts Lake Perry and passes several high bluffs. There is an unmanned water drop about 4.7 miles into this section, and 3.5 miles beyond that is the Old Military Trail trailhead, the first fully stocked manned aid station and the turnaround for the 25K course. Bald eagles may be visible in this area.

Subsequent sections include a 3.5 mile stretch along Little Slough Creek to Kiowa Road, a 3.6 mile runnable gravel segment on Kiowa Road and 82nd Street with a manned aid at the old Lowry school building, a remote 4.7 mile wooded and field section leading to the Slough Creek boat ramp aid station, and a final 6 mile northerly return to Apple Valley.
