The Gate2Gate Trail Run takes place in the northern region of San Felasco Hammock Preserve State Park just outside Alachua, Florida. The event uses a looped trail that showcases single-track mountain bike trails, open fields, rolling hills, and double-track hiking and birding trails.

After the first five miles the course moves from northern single-track into the southern park trails, follows the yellow trail loop through the western portion and passes the CSO area before returning to the northern biking trails. 25K entrants complete the circuit once; 50K entrants complete it twice. The course will be open for eight hours.

Aid stations are positioned at multiple mile markers on each loop (Loop 1: miles 5, 9, 12, 15.5; Loop 2: miles 20.5, 24.75, 28, 31.5). Stations provide water, electrolyte drink (Powerade), basic ultra foods such as PB&J sandwiches, and a selection of sweet and salty items. Gels, S‑Caps, and medications like ibuprofen will not be supplied at aid stations. Cups will not be provided; participants must carry their own bottles, hydration vests, or cups. Single‑use gel packs are discouraged to reduce trail litter.

The event is a benefit with net profits donated to Friends of San Felasco. Every finisher receives a custom medal and a custom glass; 50K finishers also receive a custom trucker hat. A TailGate2Gate celebration zone is planned for the day. The organizer policy states there are no refunds and no transfers under any circumstances. Camping options are available nearby including O'Leno State Park and Paynes Prairie Preserve State Park, each about 20 to 30 minutes from the start. A backyard ultra at San Felasco Hammock Preserve where runners complete a 4.167 mile loop each hour until one runner outlasts the rest; no aid is allowed on course and each runner has a 10 by 10 foot base camp at the start.
