Turkey Burn Ultra takes place at Lake Louisa State Park in Clermont, FL. The event uses a 16.2 mile loop course that mixes wooded hard-packed singletrack, open grass, sandy hills, and shell rock gravel roads. The park contains 11 distinct natural communities including sand hills, swamp forests, creeks, and lakes, providing varied terrain and views of Lake Louisa.

Trail composition is approximately 75% singletrack and 25% gravel or paved surfaces. The 50K course includes about 700 feet of elevation. Routes are clearly flagged and signed. The event provides four fully stocked aid stations with food, drinks, and medical support, professional timing services, a race shirt or hat, and a finisher medal. Media teams produce free race videos and photos for participants.

Timekeeping and cutoff policies include a nine hour limit for the 50K, noted as a 27 minute pace. Awards include free next-year entries for overall winners in each distance and top three awards by age class across several categories from under 25 through masters 50 and up. Race rules require completion of all course flags for credit, restrict crew access to the start/finish area, disallow pacers, and permit trekking poles. Entry transfers to another runner in the same event are allowed until five days before the event; entries are not refundable and do not transfer between events or years.
